Duke of Hamilton is a title in the Peerage of Scotland, created in April 1643. It is the senior dukedom in that Peerage, and as such its holder is the Premier Peer of Scotland, as well as being head of both the House of Hamilton and the House of Douglas. The title, the town of Hamilton in Lanarkshire, and many places around the world are named after members of the Hamilton family. The ducal family's surname, originally "Hamilton", is now "Douglas-Hamilton". Since 1711, the Dukedom has been held together with the Dukedom of Brandon in the Peerage of Great Britain, and the Dukes since that time have been styled Duke of Hamilton and Brandon, along with several other subsidiary titles.

He went with King James to Derry in 1689 and tried to negotiate the surrender of the town with Adam Murray. He raised a regiment of horse that he led in the defeats of Newtownbutler in 1689 and Aughrim in 1691. He was killed when the ship that should have brought him to France was intercepted by a Dutch privateer.

James Hamilton, 3rd Earl of Arran (1537–1609) was a Scottish nobleman and soldier who opposed the French-dominated regency during the Scottish Reformation. He was the eldest son of James Hamilton, Duke of Châtellerault, sometime regent of Scotland. He was of royal descent, and at times was third or fourth in succession to the Scottish crown; several royal marriages were proposed for him, but he eventually never married. He went to France with Mary, Queen of Scots, where he commanded the Scots Guards. After returning to Scotland, he became a leader of the Protestant party against Mary and her French supporters. However, he went insane in 1562 and was confined for the rest of his life.

Lieutenant General James Hamilton, 4th Duke of Hamilton and 1st Duke of Brandon was a Scottish nobleman, the Premier Peer of Scotland, and Keeper of the Palace of Holyroodhouse. He was a Master of the Great Wardrobe, Master-General of the Ordnance, Ambassador, and Colonel-in-Chief of his regiment. Hamilton was a major investor in the failed Darien Scheme, which cost many of Scotland's ruling class their fortunes, and he played a leading role in the events leading up to the Act of Union in 1707. He died on 15 November 1712 as the result of a celebrated duel in Hyde Park, Westminster, with Charles Mohun, 4th Baron Mohun, over a disputed inheritance.

Appointed a groom of the bedchamber to Charles II after the death of his father in battle, he took the Williamite side at the Glorious Revolution and on 21 March 1689 supplied Derry with stores that enabled the town to sustain the Siege of Derry until it was relieved in 1689. Shortly after inheriting a Scottish and Irish peerage from a second cousin, he was created a Viscount in Ireland for his services to the Williamite cause.
